The traditional way of connecting nucleic acids and antibodies mainly relies on the modification and reconnection of chemical coupling agents, such as azidating the sugar chain at the Fc end of the antibody, and then performing a coupling reaction with the DBCO-modified nucleic acid at the 5' end to complete the connection between the nucleic acid and the antibody. . This method can control the coupling position, but base modification and other purification steps are costly, and it is difficult to control the number of couplings (Figure 1).

The AND linker developed by Professor Xing Hang of Hunan University is a bifunctional fusion protein containing two functional domains (Figure 2):

Specific single-stranded DNA (ssDNA) linking domain, which can covalently bind to the sequence 5'-AAGTATTACCAGAAA-3' (underlined is the cleavage binding site) (react with the 5'-phosphate group after cleavage) );
The Fc-terminal connecting domain of the specific antibody can be stably coupled to the Fc-terminal, ultimately forming an antibody-single-stranded DNA conjugate with a stoichiometric ratio of 1:1 (Figure 3).
